Is there a way to include investment loan interest to calculate total return?

    Loan interest calculation is not supported by default.
    You can try adding negative dividends and then check results to see if that works for you.

    I tried using the MER but looks like it use the total value price in the calculation, if it use the cost basis instead then it would work well.

    No, MER cannot be used as MER is an approximate calculation based on selected period symbol end value.
